The simple difference- one is used with singular subject and the other with plural. Does goes with singular and do with plural.  She does that.  They do that.  In fact, this is the general usage. We adopt this to all the other verbs. She does sing = She sings They do play = They play

does uses for pulural and do uses for singular sentence.
